The wavelet-based adaptive large-eddy simulation method is extended for computational modelling of compressible wall-bounded attached turbulent flows. The wavelet-threshold filtered compressible Navier–Stokes equations are derived. The unclosed terms in the governing equations are approximated by using eddy-viscosity and eddy-conductivity modelling procedures based on the anisotropic minimum-dissipation approach. The proposed filtering procedure is integrated with the adaptive anisotropic wavelet collocation method, which allows for the appropriate mesh stretching in the wall-normal direction. The performance of the method is assessed by conducting adaptive numerical simulations of fully developed supersonic flow in a plane channel with isothermal walls, which represents a well-established benchmark for wall-bounded turbulent compressible flows. The present results demonstrate both the feasibility and the effectiveness of the novel wavelet-based adaptive method in the high-speed compressible regime, showing good agreement with reference numerical solutions.

Introduction
Hedonic eating refers to the consumption of food uniquely because of its </div><div>gustatory rewarding properties and not for homeostatic needs; so the subject eats also when not in a </div><div>state of energy depletion. Hedonic eating may stimulate powerfully food intake; hence, </div><div>understanding its physiological modulation could improve our knowledge of the pathophysiology of </div><div>obesity. Furthermore, obese people often present binge-eating behaviours, and binge-eating is </div><div>experienced as rewarding. Endocannabinoids, including anandamide (AEA) and 2-</div><div>arachidonoylglycerol (2-AG), and endocannabinoid-related compounds, oleoylethanolamide (OEA) </div><div>and palmitoylethanolamide (PEA), are involved in the modulation of food-related reward. </div><div>
Objectives and AimsTherefore, in the present study we aimed to investigate the responses </div><div>of endocannabinoids and endocannabinoid-related compounds to hedonic eating in obese patients </div><div>with and without binge eating disorder (BED). </div><div>
MethodsPeripheral levels of AEA, 2-AG, OEA and PEA were measured in 7 obese patients </div><div>with BED and 7 obese patients without BED after eating hedonic and non-hedonic food, and </div><div>compared to those of normal weight healthy controls. </div><div>
ResultsPeripheral levels of endocannabinoids and endocannabinoid-related compounds </div><div>exhibited different response patterns to hedonic eating among the groups. </div><div>
ConclusionsThese data confirm the role of endocannabinoids and endocannabinoid-related </div><div>compounds in food-related reward and suggest a dysregulation of their physiology in obesity and in </div><div>the BED. These findings may have potential relevance to the prevention and treatment of obesity </div><div>and BED.</div>
Attachment style and salivary cortisol awakening response in eating disorders

Introduction
Early life experiences can influence hypotalamus-pituitary-adrenal (HPA) axis regulation and adult attachment styles. Furthermore, several studies showed that in patients with eating disorders (EDs) there is a prevalence of insecure attachment. However, the relationship between adult attachment style, HPA axis functioning and onset of EDs is largely unknown.
Objectives and aimsIn order to evaluate possible associations between attachment styles and HPA axis functioning in EDs, we investigated Cortisol Awakening Response (CAR) in ED patients with different attachment styles.
MethodsTwenty adult patients with EDs were classified in three groups, according to the Experience in Close Relationship questionnaire (6 with secure attachment, 6 with anxious attachment and 8 with avoidant attachment). Saliva samples were collected at awakening and 15, 30 and 60 minutes after.
ResultsThere was a significant difference among the groups in both awakening and post-awakening cortisol concentrations. In particular, compared to secure and avoidant groups, the anxious group exhibited lower cortisol concentrations at awakening and post-awakening with a preservation of the timing of the CAR.
DiscussionPresent findings demonstrate that anxious attachment style is linked to flattened CAR in EDs. This pattern has been associated with other psychiatric disorders. Therefore, attachment style could influence the HPA functioning and it could play, although not specifically, a role in pathophysiology of EDs.

The recent development of the adaptive-anisotropic wavelet-collocation method, which incorporates the use of coordinate transforms, opens new horizons for wavelet-based simulations of wall-bounded turbulent flows. The new wavelet-based adaptive unsteady Reynolds-averaged Navier–Stokes approach for computational modelling of turbulent flows is presented. The proposed methodology that is integrated with anisotropic wavelet-based mesh refinement is demonstrated for a two-equation eddy-viscosity turbulence model. The performance of the method is assessed by conducting numerical simulations of the turbulent flow past a circular cylinder at subcritical Reynolds number. The present study demonstrates both the feasibility and the effectiveness of the new wavelet-based adaptive unsteady Reynolds-averaged turbulence modelling procedure for external flows.

The wavelet-based eddy-capturing approach with variable thresholding is extended to bluff-body flows, where the obstacle geometry is enforced through Brinkman volume penalization. The use of a spatio-temporally varying threshold allows one to perform adaptive large-eddy simulations with the prescribed fidelity on a near optimal computational mesh. The space–time evolution of the threshold variable is achieved by solving a transport equation based on the Lagrangian path-line diffusive averaging methodology. The coupled wavelet-collocation/volume-penalization approach with variable thresholding is illustrated for a turbulent incompressible flow around an isolated stationary prism with square cross-section. Wavelet-based adaptive large-eddy simulations supplied with the one-equation localized dynamic kinetic energy-based model are successfully performed at moderately high Reynolds number. The present study demonstrates that the proposed variable thresholding methodology for wavelet-based modelling of turbulent flows around solid obstacles is feasible, accurate and efficient.

A new framework for spatio-temporally adaptive turbulence simulations is proposed. The method is based on a variable-fidelity representation that tightly integrates numerics and modelling of subgrid-scale turbulence and aims to capture the flow physics on a near-optimal adaptive mesh. The integration is achieved by combining hierarchical wavelet-based computational modelling with spatially and temporally varying wavelet threshold filtering. The proposed approach provides automatic smooth transition from directly resolving all flow physics to capturing only the energetic/coherent structures, which leads to a dynamically adaptive variable-fidelity approach. The self-regulating continuous switch between different fidelity regimes is accomplished through a two-way feedback mechanism between the modelled dissipation and the local grid resolution, which is based on spatio-temporal variation of the wavelet filtering threshold. The proposed methodology systematically accounts for and exploits the spatial and temporal intermittency of turbulence. Thus, it overcomes the major limitation of all existing wavelet-based multi-resolution techniques, namely, the use of a global thresholding criterion. The procedure consists of tracking the wavelet thresholding factor within a Lagrangian frame by exploiting a Lagrangian path-line diffusive averaging approach, based on either interpolation along characteristics or direct solution of the corresponding evolution equation. This new methodology is tested for linearly forced homogeneous turbulence at different Reynolds numbers and provides very promising results on a benchmark with time-varying prescribed level of turbulence resolution.

The wavelet-based eddy capturing approach is extended to three-dimensional bluff body flows, where the flow geometry is enforced through Brinkman volume penalization. The wavelet-collocation/volume-penalization combined method is applied to the simulation of vortex shedding flow behind an isolated stationary prism with square cross-section. Wavelet-based direct numerical simulation is conducted at low supercritical Reynolds number, where the wake develops fundamental three-dimensional flow structures, while wavelet-based adaptive large-eddy simulation supplied with the one-equation localized dynamic kinetic-energy-based model is performed at moderately high Reynolds number. The present results are in general agreement with experimental findings and numerical solutions provided by classical non-adaptive methods. This study demonstrates that the proposed hybrid methodology for modelling bluff body flows is feasible, accurate and efficient.

The ability of wavelet multi-resolution analysis to detect and track the energy-containing motions that govern the dynamics of a fluid flow offers a unique hierarchical framework for modelling and simulating turbulence. In this paper, the role of the wavelet thresholding level in wavelet-based modelling and simulation of turbulent flows is systematically examined. The thresholding level controls the relative importance of resolved energetic structures and residual unresolved background flow and, thus, the achieved turbulence resolution. A fully adaptive eddy capturing approach is developed that allows variable-fidelity numerical simulations of turbulence to be performed. The new method is based on wavelet filtering with time-varying thresholding. The thresholding level automatically adapts to the desired turbulence resolution during the simulation. The filtered governing equations supplemented by a localized dynamic energy-based closure model are solved numerically using the adaptive wavelet collocation method. The approach is successfully tested in the numerical simulation of both linearly forced and freely decaying homogeneous turbulence.

The stochastic coherent adaptive large eddy simulation (SCALES) methodology is a novel approach to the numerical simulation of turbulence, where a dynamic grid adaptation strategy based on wavelet threshold filtering is utilized to solve for the most ‘energetic’ eddies. The effect of the less energetic unresolved motions is simulated by a model. Previous studies have demonstrated excellent predictive properties of the SCALES approach for decaying homogeneous turbulence. In this paper the applicability of the method is further explored for statistically steady turbulent flows by considering linearly forced homogeneous turbulence at moderate Reynolds number. A local dynamic subgrid-scale eddy viscosity model based on the definition of the kinetic energy associated with the unresolved motions is used as closure model. The governing equations for the wavelet filtered velocity field, along with the additional evolution equation for the subgrid-scale kinetic energy, are numerically solved by means of a dynamically adaptive wavelet collocation method. It is demonstrated that adaptive simulations closely match results from a reference pseudo-spectral fully de-aliased direct numerical simulation, by using only about 1% of the corresponding computational nodes. In contrast to classical non-adaptive large eddy simulation, the agreement with direct solution holds for the mean flow statistics as well as in terms of energy and enstrophy spectra up to the dissipative wavenumbers range.

EURECA (European Underground Rare Event Calorimeter Array) is an astro-particle physics facility aiming to directly detect galactic dark matter. The Laboratoire Souterrain de Modane has been selected as host laboratory. The EURECA collaboration unites CRESST, EDELWEISS and the Spanish-French experiment ROSEBUD, thus concentrating and focussing effort on cryogenic detector research in Europe into a single facility. EURECA will use a target mass of up to one ton, enough to explore WIMP – nucleon scalar scattering cross sections in the region of 10-9 – 10-10 picobarn. A major advantage of EURECA is the planned use of more than just one target material (multi target experiment for WIMP identification).

The role of coherent and incoherent subgrid-scale modes in large-eddy simulation modelling is examined. The coherent/incoherent decomposition of the subgrid-scale stresses based on the wavelet de-noising procedure is introduced. A priori dynamical tests based on the perfect modelling approach are performed for decaying isotropic turbulence. The theoretical effects of coherent and incoherent subgrid-scale forces are dynamically evaluated during the simulation. The relation between deterministic/stochastic subgrid-scale models and coherent/incoherent subgrid-scale stresses is discussed. The main result is that in large-eddy simulations low-order statistics can be almost exactly reproduced when only the effect of the coherent subgrid-scale modes is accounted for, while the incoherent subgrid-scale modes have a negligible effect upon the large-scale dynamics and the energy transfer.
Linkage disequilibrium at the cystathionine β synthase (CBS) locus and the association between genetic variation at the CBS locus and plasma levels of homocysteine

Cystathionine β synthase (CBS) is a key enzyme in homocysteine metabolism. We have examined four apparently non-functional polymorphisms in the CBS gene and have determined their frequency, degree of linkage disequilibrium and association with plasma homocysteine levels. The polymorphisms are a 68 bp insertion in exon 8, C699T in exon 8, C1080T in exon 11 and C1985T in the 3′ untranslated region. 785 individuals participating in the European Atherosclerosis Research Study II (EARSII), from 11 countries across Europe were genotyped for these polymorphisms. The 68bp insertion had the highest frequency in the UK and in the Middle region, with a lower frequency in the Baltic and the South (p=0.01), and the exon 11 polymorphism had the highest frequencies of the rare allele in the Baltic (p<0.05). There was a high degree of linkage disequilibrium between the polymorphisms (p<0.001 overall), except between C699T and the C1985T, with three common haplotypes accounting for nearly 80% of chromosomes. Examination of the association between these polymorphisms and plasma homocysteine levels revealed that the carriers of the rare alleles of the C699T, C1080T and C1985T polymorphisms had lower plasma homocysteine concentrations than those homozygous for the common alleles, although these differences were not statistically significant. The thermolabile valine variant caused by a substitution of a C for a T at nucleotide 677 in the methylenetetrahydrofolate reductase (MTHFR) has previously been shown to have profound effects on plasma levels of homocysteine in this sample, but the homocysteine-raising effect associated with this thermolabile variant was not seen in carriers of the 68 bp insertion, with this interaction being statistically significant (p<0.001). These data demonstrate that variation in the CBS gene as detected with these four polymorphisms, had no statistically significant effect on plasma homocysteine levels in these healthy young men. However, the presence of the 68 bp insertion, which is found in approximately 7.5% of individuals in the populations of Europe sampled, abolishes the raising effect of thermolabile MTHFR Val/Val genotype, and may be of importance in the situation of high homocysteine.
